An oxidoreductase is highly expressed in a virulence - loss - mutant of the cabbage yellows fungus having biocontrol activity

Abstract

Cong:1-2 is a virulence-loss-mutant of the soilborne cabbage yellows fungus ( Fusarium oxysporum f. sp. conglutinans; isolate Cong:1-1) emerged during repeated passages. Pre-treatment of cabbage seedlings with Cong:1-2 showed reduction of yellows, suggesting that Cong:1-2 carried biocontrol activity.1, 2) 2D electrophoresis showed a protein ( pI ~5.2, ~75 kD ) highly expressed especially in Cong:1-2. This protein was presumed as a GMC oxidoreductase by BLAST analysis. The gene encoding this protein in Cong:1-2 was cloned, and the putative ORF ( 1974 bp ) composed of two exons was named odx1. The ~5-kb genomic sequences including odx1 showed 100% homology between Cong:1-1 and Cong:1-2. To examine the relationships between ODX1 and pathogenicity or biocontrol activity, we generated odx1-disruptants from Cong:1-2.
